The Effects of Rho Kinase Inhibitor Y 27632 on Morphological Changes of Trabecular Meshwork in Juvenile Rats Model with Sodium Hyaluronate Injection

Objective/Purpose
To study the effect of rho kinase inhibitor Y 27632 on schlemm's canal diameter, trabecular meshwork thickness, and intercellular space of juxtacanalicular meshwork in juvenile rats model with sodium hyaluronate injection

Materials/Patients
Twenty four juvenile Wistar rats aged 4-6 weeks , weighing 100-150 g were used in this study. The rats were sacrificed after receiving treatment of sodium hyaluronate and Y 27632 and then the eyeballs were enucleated by dissecting the extraocular muscles.

Methods
An experimental study with post test only control group design. The samples were divided into 6 groups , negative control, positive control I with intracameral injection of sodium hyaluronate, positive control II with topical Y 27632 10 mM, and three experimental groups with intracameral injection of sodium hyaluronate and Y 27632 0.1 mM, 1 mM, and 10 mM. Morphological changes evaluation included schlemm's canal diameter, trabecular meshwork thickness, and intercellular space of juxtacanalicular meshwork. Quantitive measurements were taken using computerized image analysis with dot slide program. Statistical analysis of one way ANOVA and independent t-test we used in this study.

Results and Conclusion
There were significant differences statistically between control and experimental groups in all three variables measurements (p-value<0.05). The highest mean of increasing schlemm's canal diameter, decreasing of trabecular meshwork thickness, and increasing of intercellular space of juxtacanalicular meshwork were noted in the experimental group given sodium hyaluronate and Y 27632 10 mM with values of 118.42002 μm, 41.2500 μm, and 2.1525 µm respectively.
There were increasing diameter of schlemm's canal, decreasing thickness of trabecular meshwork, and increasing of intercellular space of juxtacanalicular meshwork due to the effect of rho kinase inhibitor Y 27632 in juvenile rats with sodium hyaluronate injection
