The Pupil Response to Light in Healthy Human Eyes by a Novel Infrared Pupillometer

Objective/Purpose
Using a new wearable infrared pupillometer to evaluate the pupil responses in human eyes.

Materials/Patients
38 eyes of healthy human (6 males and 32 females with age (20-50 y/o), visual acuity ≧ 0.8, and refraction error (+3~-3D)); six eyes were excluded.

Methods
This study uses the pupillometer to observe the pupil response of different light stimulus in dark room and evaluate the maximum constriction velocity and restoration velocity. The different wavelength and intensity of light sources are shown as the following: (1) White light, (2) Red light (wavelength = 640nm), (3) Green light (wavelength = 534nm), and (4) Blue light (wavelength = 470 nm). The intensity are 500 mcd and 50 mcd. Resting pupil diameters, duration of pupil constriction and restoration to 75% pupil size, and their maximum constriction and restoration velocities were measured.

Results and Conclusion
The mean resting pupil diameter/iris diameter in dark is 54.28±5.39%. The difference (%) between resting size and the constricted size after 50 mcd (W, R, G, B) light stimulation are 11.0±3.2, 12.41±3.6, 12.82±4.77, 13.08±3.36, respectively. The difference after 500 mcd light stimulation are 12.64±7.39, 14.13±7.5, 13.50±2.52, 13.21±3.25. The duration of resting pupil constricting to the minimum after 50 mcd light stimulation are 0.91±0.1, 0.92±0.06, 0.93±0.08, 0.95±0.09 sec. The duration after 500 mcd light stimulation are 0.91±0.22, 0.93±0.15, 0.97±0.08, 1.04±0.31 sec. The duration between the constricted size and restoring to the 75% pupil size after 50 mcd light stimulation are 1.83±0.58, 2.02±1.26, 1.92±0.65, 1.75±0.45 sec. The duration after 500 mcd light stimulation are 2.11±0.87, 2.02±0.63, 2.27±0.97, 2.12±0.77 sec. The maximum pupil constriction velocities (MPCV) after 50 mcd (W, R, G, B) light stimulation are 1.86±0.43, 1.88±0.43, 2.02±0.45, 2.02±0.46 %/sec, respectively. MPCV after 500 mcd (W, R, G, B) light stimulation are 1.90±0.42, 1.88±0.42, 1.94±0.44, 2.06±0.41 %/sec, respectively. The maximum pupil restoration velocities (MPRV) after 50 mcd (W, R, G, B) light stimulation are 0.87±0.27, 0.93±0.40, 0.89±0.25, 0.89±0.21 %/sec, respectively. MPRV after 500 mcd (W, R, G, B) light stimulation are 0.88±0.27, 0.87±0.26, 0.85±0.2, 0.89±0.33 %/sec, respectively. The different wavelengths and intensities make no significant changes of MPCV and MPRV.
We found the MPCV and MPRV of pupil responses are not changed by different wavelengths (W, R, G, B) light and different intensity (50, 500 mcd) in the study.
