RP58 is a transcriptional repressor which plays a significant roles during corticogenesis. It is known that RP58 promoter is directly activated by Ngn2 in the process of the neuronal differentiation. Recently it has been reported that Hes1 and Ngn2 expressions dynamically oscillate in neural progenitor cells to maintain their undifferentiated state. Since RP58 is activated by Ngn2, it is possible that RP58 is also expressed oscillatory manner by reacting on Ngn2 expression in neural progenitors. To examine RP58 gene promoter activity in neural progenitors, we performed real-time imaging using ubiquitinated firefly luciferase reporter conjugated to 5 kb RP58 promoter. Neural progenitor cells prepared from mouse embryonic telencephalon were electoroporated with RP58 promoter-Ub-luc constructs and luciferase activities were visualized by luminescence live cell imaging system in stem cell maintenance medium. As a result, oscillatory expression of luciferase driven by RP58 promoter was observed.
