Generation of Immunotoxins against AML: Cytocidal activities with anti-CD71 mAb

We developed a unique screening system to establish cancer-targetable antigens/antibodies sets, which contained the catalytic and translocation domain of the truncated diphtheria toxin (DT) as well as the three IgG-binding C domains of Streptococcal protein G (3C). Mice were immunized with a AML line, TF-1, and hybridoma libraries were prepared. In total 69 mAb were obtained which performed highly efficient of AML cells. In clear contrast, the selected few among our DT3C-targeting mAb, including anti-CD71, anti-CD43, and anti-CD99, when conjugated with DT3C, revealed remarkable iTox cytotoxicity on TF-1 cells. Toxin conjugates with anti-CD71 mAb showed highly augmented cytocidal activities against all the AML cells tested, leading to a complete cell death; cytotoxicities as revealed by a 72hr WST-1 assay resulted in 79.0% and 80.8%, for TF-1 and SKM1 cells, respectively. Using Immunotoxins that super-targeting mAb would be promising for the treatment of AML.
