IFN-γ-mediated elimination of host DCs by cytokine induced killer cells leads to less GVHD

Cytokine-induced killer (CIK) cells are ex vivo-expanded T lymphocytes expressing both natural killer (NK)- and T-cell markers. We have reported that adoptive transfer of allogeneic CIK cells in a murine model caused minimal graft-versus-host disease (GVHD) with retention of antitumor activity mediated by NKG2D, which is an activating receptor expressed on NK cells. The mechanism of suppression of GVHD with CIK cells is not fully understood. First we clearly demonstrated that CIK cells had strong killing activity against allogeneic dendritic cells (DCs), which were generated from rodent bone marrow cells, by 51-Cr release cytotoxicity assays. In addition, killing activity against DCs did not changed with/without adding NKG2D blocking antibody, suggesting that there were possible mechanisms for cell killing other than NKG2D/NKG2D ligand system in CIK cells. Next we have shown that the residual number of host-typed splenic DCs in the mice receiving bone marrow (BM) cells plus CIK cells was significantly lower compared to the mice receiving BM plus spleocytes to induce GVHD (p<0.05). Finally as previous report showed that CIK cells generated from IFN-γ knock out mice developed severe GVHD, we compared killing activity against allogeneic DCs between WT and IFN-γ KO CIK cells. As expected, killing activity of IFN-γ KO CIK cells was much less compared to that of WT CIK cells (p <0.05). In conclusion, allogeneic CIK cells caused less GVHD due in part to elimination of host DCs mediated by IFN-γ.
