CBFA2T3-GLIS2 fusion gene defines an subtype of pediatric acute megakaryoblastic leukemia

Backgrounds: Although acute megakaryoblastic leukemia (AMKL) patients with Down syndrome (DS) have excellent survival, non DS-AMKL patients have an extremely poor outcome. Recently the CBFA2T3-GLIS2 fusion gene was reported as a strong prognostic factor in pediatric AMKL (Cancer Cell 2012). Therefore, we performed RT-PCR for CBFA2T3-GLIS2 on leukemic blasts at the time of diagnosis from 5 pediatric patients with AMKL at our hospital.Materials and Methods: Between 2003 and 2012, peripheral blood mononuclear cells or bone marrow with leukemic blasts were collected from 5 patients with AMKL at our hospital (nonDS-AMKL n=3, DS-AMKL n=2). We retrospectively analyzed the fusion gene, CBFA2T3-GLIS2 in all 5 samples by RT-PCR or direct sequence.Results: We identified the CBFA2T3-GLIS2 fusion gene transcript in one patient with non-DS AMKL. All patients achieved complete hematological response after induction therapy. Four patients who did not carry the CBFA2T3-GLIS2 fusion gene transcript are in complete remission. Whereas, the patient who carried CBFA2T3-GLIS2 fusion gene transcript died after hematopoietic stem cell transplantation from AMKL relapse. Conclusion: The results of presented cases suggest that the fusion gene, CBFA2T3-GLIS2 is also related to non-DS AMKL in Japanese pediatric population and may be a prognostic factor in patients with AMKL.
